Multipoint confocal microscope with a module for laser ablation
The device was bought with funds from the ERC project NewSpindleForce and can be used only within the project until March 31st, 2020. Inverse fluorescence scanning microscope • Motorized 6-position emission filter wheel; Custom dichroics and polychroics for multiple excitation/emission wavelengths; fluorescence filters for DAPI, GFP and RFP; laser ekscitation lines for DAPI, GFP i RFP. • Motorized head with objectives: 20×, 100×/1.4. • Combination galvanometer and piezoelectric crystal scanning (Z-stack). • Fully motorized aperture plate with 7 software-selectable apertures; 3 pinhole settings (30, 45, and 60 μm); 4 slit settings (22, 35, 50, and 70 μm). • Speed 1000 images per second, pixel size 64x64. Possibility of simultaneous acquisition in three detection channels. Laser ablation • Ablation laser: puls shorter than 300 fs, wavelenghth between 800 i 1064 nm, minimal power 1 W, frequency 10-80 MHz • Precise ablation of areas wide around 500 nm • EMCCD camera 1000 pictures with 64x64 pixel size (binning 1x1) • Camera pixel size up to 16 µm • Image pixel size less than 100 nm • Automatic focus
